Book excerpt: With the increased practice of testing oral fluid for the detection of illicit drug use, the available active and passive collection methods have also increased. Active collection methods stimulate oral fluid production of the salivary gland through acidic or non-acidic methods. In contrast, passive collection methods simply use a collection device that is held in the oral cavity to accumulate oral fluid. This study focused on the recovery of drug analytes from solutions used in active and passive collection methods after experiencing the same solid-phase extraction method. A total of eleven drug analytes were examined including benzodiazepines, cocaine, methamphetamines, opioids, and delta-9-tetrahydrocannabinol (THC). Recovery samples were analyzed via Gas Chromatography–Mass Spectrometry (GC/MS). Of the eleven compounds tested during the recovery study, five were found to have statistically significant differences between the percent recoveries in active and passive oral fluid solutions. The active collection solution yielded higher percent recoveries than the passive collection solution. These findings suggest that active solution would be a more viable collection method to recover a greater majority of the drug analyte present in oral fluid.
